Mutation of the Meishan-specific GATA-4 binding site diminished promoter activity in αT3-1 cells. Cells were transiently transfected with luciferase (LUC) vectors containing either the native, full-length Meishan promoter (−M5118pGL3), a block replacement mutation of the GATA-4 binding site within the context of −M5118pGL3 (−MμGATA4pGL3), or promoterless control (pGL3).
