AQP5 mRNA and protein expression in porcine myometrial explants. The expression of AQP5 mRNA in the myometrial tissue harvested from pigs (n = 5) on Days 10–12 (a) and 14–16 (b) of the estrous cycle after treatment with steroids (P4, E2), oxytocin (OT), arachodonic acid (AA), forskolin (FSK) and cAMP for 3 and 24 h. The expression of the studied gene, determined by real-time PCR, is presented as means ± SEM in relation to the expression of reference genes, GAPDH and 18 sRNA. Different letters indicate significant differences (p <0.05) between each treatment and respective control values for 3- (a, b) or 24-h (c, d) incubations. The protein content of AQP5 was determined by semi-quantitative Western blot analysis, in the myometrial explants harvested from pigs (n = 5) on Days 10–12 (c) and 14–16 (d) of the cycle after treatment with the abovementioned factors for 3 and 24 h. The data is presented as means ± SEM in relation to β-actin tissue content. Different letters indicate significant differences (p <0.05) between each treatment and respective control values for 3- (a, b) or 24-h (c, d) incubations.
