Binding profile of recombinant human ZP1 273-551aa with human spermatozoa. Capacitated/acrosome-reacted sperm (5 × 106/50 μl) were incubated with 2.5 mg of FITC conjugated baculovirus-expressed ZP1273-551aa and processed as described in Methods. Panels (A) and (B) represent the binding of FITC conjugated ZP1273-551aa with capacitated acrosome intact sperm whereas Panel (C) depicts the binding profile with acrosome-reacted spermatozoa. The acrosomal status was determined by labeling the sperm with TRITC-PSA. The images were captured using Eclipse 80i fluorescence microscope (Nikon). In each panel, the sub-panels are represented as a: phase contrast; b: TRITC-PSA fluorescence; c: FITC-ZP protein fluorescence and d: overlap of fluorescent frames. The scale bar represents 2.5 μm.
