Detection of IP 3 R isoforms in mouse ovarian tisuue. Ovarian tissue was incubated with specific antibodies against IP3R types 1, 2, and 3, as well as with TO-PRO-1 iodide (50 nM) to label nuclei. Control images done in absence of primary antibody are at the top of columns 1 and 2. The first column shows entire ovarian structures, whereas the second column depicts an enhanced image delimiting granulosa (G) and theca (T) cells (Magenta color). The third column shows nuclear signal associated with TO-PRO-1 (Cyan color), and the fourth column represents the convergence of signals between IP3Rs and nuclear structures (merge). Scale bar for ovary slices corresponds to 100 μm, and for granulosa cells to 20 μm. These results provide evidence for the intranuclear presence of all the IP3R isoforms in the mouse granulosa cells in situ. Representative of 3 independent experiments.
