Anion-exchange chromatography (AEC) of FPE. Approximately 40 pituitaries were extracted twice with 0.5 ml 50 mM Tris-HCl (pH 7.5) and the extracts were combined, centrifugally filtered, and applied to a Mono Q column via the 10-ml Superloop. The effluent was monitored for UV-absorbance (A) and a gradient of NaCl was applied after non-adsorbing material percolated through the column. Two-ml fractions were collected and tested for (B) maturational and (C) steroidogenic activity. For this experiment only, positive controls (FPE added at a concentration of 0.5 pit. equiv./well) were 43% GVBD with DHP-, T-, and E2-secretion of 0.68, 0.54, and 3.4 ng/well, respectively.
